After that the VMs will be recognized as same as other hosts that also connect to the real SBS network and configure the port mapping on router to the private address we assigned to that VM. So could we first enabling the UPnP feature on our edge router if that is supported and let SBS server automatically configures these settings by the built-in wizards?

We ran SW in a VM without a problem for about a year we just recently moved it. This strikes me as the "when in doubt, do this" answer.
